Detection and Characterization of Hepatitis C virus in Korean non-A, non-B Hepatitis Patients

Abstract

|
To characterize hepatitis C virus stains present in Korean non-A, non-B hepatitis, RNA was extracted from patients' sera using guanidium thiocyanate-phenol-chloroform mixture. RNA was then examined with a combined assay of reverse transcription
and
polymerase chain reaction, using 4 different primer pairs derived from conserved regions in 5' -NCR, and putative core, E2/NS1, NS3 region of the isolates from the United States, Japan and France.
Among 147 cases, hepatitis C virus RNA could be detected from 106(72.1%) cases using 5' -NCR primer; whereas positive rates againt core, E2/NS1. NS3 primers were 46(31.3%), 27(18.4%), 61(41.5%), respectively.
We can divide these 106 cases into 6 patterns based upon reactivity against each 4 primer pairs Among these 106 cases, 29(27.4%) show reactivity against 5' -NCR primer only. These findings indicate that there are considerable heterogeneity in
hepatitis
C virus genomes in Korea.
